TB-500 is a synthetic variant of the naturally occurring peptide found in nearly all human and animal cells, Thymosin Beta 4 (TB500). It is a pioneering drug candidate that facilitates the following: differentiation of endothelial (blood vessel) cells, angiogenesis (the formation of new blood vessels from existing ones) within dermal tissues, keratinocyte migration, collagen deposition, and a reduction in inflammation.
A primary mechanism of TB500 is its capacity to regulate the cell-structural protein, Actin, which is essential for cell architecture and movement. Among the myriad of proteins found within cells, actin can account for up to 10% of the total protein content, thereby playing a crucial role in the cell's genetic framework.
This powerful peptide is part of a widespread family of 16 related molecules that exhibit significant conservation of sequence and localization across most tissues and circulating cells. TB500 not only interacts with actin but also inhibits actin polymerization, acting as the actin-sequestering molecule in eukaryotic cells.
TB500 was identified as a gene that is up-regulated by four to six times during the initial stages of blood vessel development and is known to encourage the generation of new blood cells from existing vessels. This peptide is found in wound fluid, and when injected subcutaneously, it enhances wound healing, muscle growth, and accelerates recovery times for muscle fibers and cells.
Another important role of TB500 is its ability to facilitate cell migration through a specific interaction with actin within the cell cytoskeleton. Research has shown that a central small amino acid long-actin binding domain possesses both blood cell reproduction and wound healing properties. These attributes are revealed through the enhanced migration of endothelial cells and keratinocytes, as well as increased production of enzymes that degrade the extracellular matrix.
Studies indicate that TB500 is a powerful, naturally occurring factor in wound repair with anti-inflammatory effects. Unlike other repair factors, such as growth factors, TB500 encourages the migration of endothelial cells and keratinocytes, does not bind to the extracellular matrix, and has a low molecular weight, allowing it to traverse considerable distances through tissues.
